The antibody variable region genes (VH and VL) from a given antibody can be reformatted to encode different constant regions, including Fab. In addition to changing the Fc portion of antibodies, the variable regions of immunoglobulins can also be reformatted into various fragments, such as Fab, F(ab)' 2, scFv, VHH and small antibodies, or can be combined with a bispecific secondary antibody. Smaller antibody formats can be used to improve tumor/tissue permeability and are relatively easy to express in non-mammalian systems.
