Amyloid beta precursor protein (APP) is a complete membrane protein expressed in many tissues and concentrated in neuronal synapses. It functions as a cell surface receptor and considered to be a modulator of synapse formation, neuroplasticity, antibacterial activity, and iron output. It is encoded by the gene APP and regulated by substrate presentation. APP is best known for its precursor molecule proteolysis to produce amyloid β (Aβ), which is a polypeptide containing 37 to 49 amino acid residues, and its amyloid fibrous form is the main component of amyloid plaques in patients with Alzheimer’s.

According to the "amyloid hypothesis", missense mutations in the APP gene promote the production of Aβ by facilitating the proteolytic processing of APP by β-or γ -secretase. In addition, the APP mutation within the A β sequence enhanced the self-aggregation of Aβ to amyloid fibrils. The cloning of presenilin (PS) protein and the mutations in PS1 and PS2 that cause AD also promote the processing of APP to form amyloid Aβ.
