Weighted UniFrac Analysis

What is Weighted UniFrac Analysis?

UniFrac analysis uses the evolutionary information between the sequence of each sample to compare whether environmental samples have significant differences in microbial communities in a specific evolutionary lineage. UniFrac can be used to evaluate and analyze the beta diversity, that is, to compare and analyze the samples to obtain the unifrac distance matrix between the samples. From the definition of UniFrac, it can be seen that it only considers whether the sequence appears in the community, and does not consider the abundance of the sequence. If the two communities contain exactly the same species, regardless of whether there is a difference in the abundance of each species or the magnitude of the difference, the UniFrac value is 0.

Overall solutions

CD ComputaBio offers the following services to meet the specific needs of different customers:

  • Multi-sample similarity tree analysis based on UniFrac

CD ComputaBio first use the Unifrac analysis method, the distance matrix obtained can be used in a variety of analysis methods, through the hierarchical clustering (Hierarchical cluatering) non-weighted group average method UPGMA to construct the evolutionary tree and other graphical visualization processing, which can visually display the microorganisms in different environmental samples Evolutionary similarities and differences.

  • PCoa analysis based on UniFrac

The distance matrix obtained by Unifrac analysis can be used in a variety of analysis methods. It can be analyzed by PCoA, a multivariate statistical method, to visually display the similarities and differences in the evolution of microorganisms in different environmental samples.

Software and algorithm

FastTree was used to build evolutionary trees according to the maximum likelihood method (approximately-maximum-likelihood phylogenetic trees).

Fastunifrac was used to analyze the distance matrix between samples.
